报告简介:

Multiprotein complexes regulate virtually all fundamental cellular activities. However, many of these essential machines exist in very low endogenous amount in cells (particularly in eukaryotes), which makes it very difficult and laborious to extract enough multiprotein complexes of sufficient purity from native sources for detailed structural and functional studies. Therefore, recombinant production is required to address this bottleneck. In order to streamline the recombinant production of multiprotein complexes, an array of advanced expression systems (termed “Multiexpression Technologies”) have been developed in the Berger laboratory (EMBL Grenoble) for various host organisms including E. coli, insect cells and mammalian cells. Those systems, especially the MultiBac baculovirus/insect cell system, have greatly contributed to structural and functional studies of numerous important multiprotein complexes at molecular and atomic details. Notably, this includes also the human general transcription factor TFIID (~1.5 MDa, 14 subunits), which plays a vital role during the initiation of eukaryotic transcription. My contributions to the expression technology development and the structural elucidation of human TFIID complexes will be discussed in details in this talk.
