报告简介:

Synaptic plasticity is the cellular model for learning, memory and other experience-dependent brain functions; its malfunction will lead to brain diseases like in Alzheimer’s disease and mental retardation etc. NMDA receptor-dependent long term potentiation (LTP) and long term depression (LTD) are two major forms of synaptic plasticity and protein phosphatase-1 (PP1) regulates the induction threshold of LTP and LTD, favoring LTD induction. PP1 has been further shown to be a molecule of forgetfulness in a mice model. Despite PP1’s critical function in synaptic plasticity and memory formation, how PP1 is regulated in these processes remains unclear.

We discovered that PP1 needs targeting, via interaction with neurabin, for LTD induction. However, spinophilin, another major synaptic PP1 binding protein does not support PP1’s function in LTD induction, elucidating the exquisite spatial requirement for PP1 targeting within spines in synaptic plasticity. Moreover, we have found that PP1 inhibitor-2, instead of inhibitor-1, is critical for LTD induction and memory formation. Interestingly, these functional studies indicate that inhibitor-2, despite its name, is a positive regulator of PP1. Future work will be directed to unravel the molecular mechanism/signaling pathways underlying inhibitor-2’s positive regulation of PP1 function in vivo.
